В исследовании была использована коллекция образцов от 82 ВИЧ-инфицированных лиц, проживающих в Алтайском крае, собранная с их информированного согласия в 2017 г. Нуклеотидные последовательности области гена pol, кодирующей протеазу и часть обратной транскриптазы ВИЧ-1, получали методом in house путем прямого секвенирования амплифицированных фрагментов гена pol. Генотипирование, филогенетический и рекомбинантный анализ проводили с использованием программ HIVdbProgram: Sequence Analysis, COMET HIV-1, REGA HIV-1 Subtyping Tool (V3), MEGA 5.05, RIP и jpHMM.</p><p>Результаты и их обсуждение. Показано, что в Алтайском крае доминирующим генотипом в регионе на 2017 г. (период сбора образцов) была циркулирующая рекомбинантная форма CRF63_02A1 (61%), суб-субтип А6 составлял 33%, на остальные варианты, такие как В, G, URF, пришлось 6%. По данным филогенетического анализа нуклеотидных последовательностей, рекомбинантная форма CRF63_02A1 аналогична таковым, циркулирующим в Сибири и на Дальнем Востоке России. Все штаммы ВИЧ-1 субтипа А принадлежат варианту А6, доминирующему в России. Анализ с применением программы RIP позволил выявить три уникальные рекомбинантные формы (URF), образованные вариантами CRF63_02A1 и А6. Мутации лекарственной устойчивости были зарегистрированы у 8 из 21 пациента (8/21, 38%), получающих антиретровирусную терапию. Условный показатель первичной лекарственной устойчивости в популяции составил 5,1%.</p></sec><sec><title>Заключение</title><p>Заключение. В настоящее время происходит процесс смены доминирующего штамма на CRF63_02A1 в Алтайском крае, где еще 13 лет назад основным был вариант ВИЧ-1 А6 (IDU-А).</p></sec></abstract><trans-abstract xml:lang="en"><p>IThe aim of this study was to characterize HIV-1 genetic strains currently circulating in Altay Kray (Western Siberia) and to analyze the HIV resistance on this territory.</p><sec><title>Materials and methods</title><p>Materials and methods. Blood samples were collected, with informed consent, in 2017 from 82 HIV infected persons living in Altai Kray. Sequences of pol gene fragments coding protease and part of reverse transcriptase were obtained by in house system and Sanger sequencing. Genotyping, phylogenetic and recombinant analyses were carried out by HIVdbProgram: Sequence Analysis, COMET HIV-1, REGA HIV-1 Subtyping Tool (V 3.0), MEGA 5.05, RIP and jpHMM.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. The results of genotype analysis revealed that the circulating recombinant form CRF63_02A1 dominated in Altay Kray (61%), subtype А was identiﬁed in 33%, the remaining subtypes, such as B, G, URF, accounted for 6%. According to phylogenetic analysis results, CRF63_02A1 sequences formed the common branch with nucleotide sequences of strains found in other regions of Siberia and Far East. All of HIV-1 variants belonging to subtype A clustered together with nucleotide sequences of A6 dominating in Russia. RIP analysis allowed to identify three unique recombinant forms (URFs), formed by CRF63_02A1 and A6. Drug resistance mutations were identiﬁed in 8 of 21 ART patients (8/21, 38%). The prevalence of drug resistance mutations in naïve patients equaled to 5,1%.</p></sec><sec><title> Conclusion</title><p> Conclusion.
