Сравнение генетического разнообразия белка Rev ВИЧ-1 суб-субтипа А6 у людей, живущих с ВИЧ (ЛЖВ), c разными стадиями заболевания.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Анализировались клинические образцы цельной крови, полученные от 227 ЛЖВ, ранее не получавших лечения, которые наблюдались в ГКУЗ Московской области «Центр по профилактике и борьбе со СПИДом и инфекционными заболеваниями». Проводили экстракцию провирусной ДНК, амплификацию области первого и второго экзона гена rev с последующим секвенированием продуктов амплификации. Генотипирование полученных последовательностей выполняли путем анализа второго экзона гена rev. Затем проводили сравнение консервативности и аминокислотных замен в последовательностях белка Rev у пациентов из различных групп.</p><p>Результаты и их обсуждение. 220 из 227 (96,9%) образцов содержали ВИЧ-1 суб-субтипа А6. У пациентов с IV стадией консервативность в белке Rev была меньше. Обнаружены две замены (L13I, S113P), которые встречались со статистически значимой разницей у ЛЖВ с разными стадиями ВИЧ-инфекции.</p></sec><sec><title>Заключение</title><p>Заключение. Полученные результаты подтверждают гипотезу о корреляции между аминокислотной последовательностью белка Rev и течением ВИЧ-инфекции и обозначают необходимость дальнейших исследований этого вопроса.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>The aim of the study</title><p>The aim of the study. The comparison of genetic diversity of Rev protein in HIV-1 sub-subtype A6 in people living with HIV (PLHIV) with different stages of the disease.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. 227 whole blood clinical samples received from PLHIV who have not had previously treatment and observed in Moscow Regional Center for the Prevention and Control of AIDS and Infectious Diseases were analyzed. The extraction of proviral DNA, the ampliﬁcation of the ﬁrst and the second rev exons with the followed sequencing was carried out. Subtyping was conducted by analyzing the rev second exon. Then the conservatism and amino acid substitutions in Rev sequences were compared in patients from different groups.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. 220/227 (96.9%) samples contained HIV-1 sub-subtype A6. In patients with IV stage of the disease the conservation in Rev was signiﬁcantly lower. There were found two substitutions (L13I, S113P) with a statistically signiﬁcant difference in occurrence in PLHIV with different stages of HIV infection.</p></sec><sec><title>Conclusion</title><p>Conclusion.
